Wilson’s disease is a rare autosomal recessive genetic disorder caused due to abnormal accumulation of copper in the liver, brain and other tissues due to defect in ATP7B gene. Early detection of excess copper levels is extremely important to prevent hepatic and neurological damage. In this project, an innovative, simple, low-cost and non-invasive method is proposed to detect Wilson’s disease through saliva sample using potassium ferrocyanide based strip technique. Potassium ferrocyanide specifically reacts with excess copper ions present in saliva and produces visible color change. Neutral buffer is added to maintain stable pH for accurate reaction kinetics, while distilled water is used as a control. In this study, strips are prepared by impregnating reagent mixture and allowed to react with saliva samples. The colour change produced is analysed and compared with control to identify the presence of elevated copper levels. This method avoids blood sample requirement, reduces cost and can be performed even in basic laboratory or field level conditions. This type of proposed strip based screening approach can be useful for preliminary identification of Wilson’s disease patients at early stages, especially in rural and resource limited settings, and also supports further confirmation using advanced diagnostic tests.
